Western Blot Test
1. HEK293 cell transfected with pcDNA alone and pcDNA-ARRB2 (Molecular weight is ~50 KDa).
2. Each lane was loaded with 10 µg total cell lysate.
3. This working concentration of the antibody is 2.75 µg/mL.

Storage
It is supplied as affinity purified antibody/PBS bufferin lyophilized form. Redissolve the powder with100µL sterile water will restore to the originalconcentration at 1 mg/mL. Store at 4oC for short-term application. For long-term storage, aliquot andstore at -20oC.

Target description
Members of arrestin / beta-arrestin protein familyare thought to participate in agonist-mediateddesensitization of G-protein-coupled receptors andcause specific dampening of cellular responses tostimuli such as hormones, neurotransmitters, orsensory signals. Arrestin beta 2, like arrestin beta 1,was shown to inhibit beta-adrenergic receptorfunction in vitro. It is expressed at high levels in thecentral nervous system and may play a role in theregulation of synaptic receptors. Besides the brain, acDNA for arrestin beta 2 was isolated from thyroidgland, and thus it may also be involved in hormone-specific desensitization of TSH receptors. Multiplealternatively spliced transcript variants have beenfound for this gene, but the full-length nature ofsome variants has not been defined.

Antigen
This polyclonal antibody was raised byimmunizing rabbit with a synthetic peptidelocated on the putative C-terminal domain(a.a. 195-350) of human Arrestin beta 2(ARRB2) (ACCESSION # BAD93023). Thepeptide immunogen is conserved with the ratand mouse counterparts.

Application
The antibody titer is more than 1000K forELISA and tested positive for Western blotanalysis of transfected total cell lysate. It hasnot been tested in the other applications.However, for the first testing, we recommend1/5,000 dilution for ELISA, 1/1,000 dilution forWestern blot analysis (WB) of recombinantprotein, 1/400 dilution for tissue extracts orcell lysates, 1/100 dilution for immuno-histochemistry (IHC) staining on frozencryosections, 1/50 dilution for IHC staining onparaffin embedded sections.
