NORDENZYME™ is the name of a group of polyclonal immunologic reagents to enzymes.

Two different types of rabbit or sheep anti-enzyme products are offered: the isolated IgG

fractions (7S) of the specific antisera and the isolated antibodies (PAb) to the enzymes.

Products against some 180 enzymes are available either as 7S or PAb or in both forms. The

7S fractions are also available in biotinylated form.

The NORDENZYME™ products have been obtained by immunization with highly purified

enzymes.

Description Biotin-conjugated IgG fraction of polyclonal rabbit antiserum to 3-phosphoglyceric

phosphokinase from baker’s yeast

Physical form Biotin-coupled hyperimmune rabbit IgG lyophilised from a solution in phosphate buffered

saline (PBS, pH 7.2).

Preservative No preservative added, as it may interfere with the antibody activity. No foreign protein added.

Immunogen 3-Phosphoglyceric phosphokinase isolated and purified from baker’s yeast. Freund’s complete

adjuvant is used in the first step of the immunization procedure.

Purification The IgG (7S) fraction is prepared from the antiserum by ammonium sulphate precipitation and

ion exchange chromatography.

Identity & Specificity The reagents were evaluated for potency, purity and specificity using most or all of the

following techniques: immunoelectrophoresis, cross-immunoelectrophoresis, single radial

immunodiffusion (Ouchterlony), block titration, ELISA, immunoblotting and enzyme inhibition.

Cross-reactivity Cross-reactivities against enzymes of other sources may occur but have not been determined.

Physicochemical

characteristics

IgG protein concentration 10 mg/ml. Biotin/ IgG protein molar ratio (B/P) approximately 6.1. No

foreign proteins added.

Marker N-Hydroxysuccinimidobiotin.

Conjugation procedure A proprietary technique for the binding to biotin is used, followed by several purification steps.

After each step activity and specificity are tested in a variety of techniques. The conjugate is

lyophilised to assure stability and long shelf life.

Intended use This product is intended for use in precipitating and non-precipitating antibody-binding assays

(such as e.g., ELISA and Western blotting and immunofluorescence or histochemical

techniques).

Directions for use Working dilutions in non-precipitating antibody-binding techniques may vary widely, but may

be between 1/1,000 and 1/30,000.

Handling The lyophilised conjugate is shipped at ambient temperature and may be stored at +4°C;

prolonged storage at or below -20°C. It is reconstituted by adding 1.0 ml sterile distilled water,

spun down to remove insoluble particles, divided into small aliquots, frozen and stored at or

below -20°C. Prior to use, an aliquot is thawed slowly at ambient temperature, spun down

again and used to prepare working dilutions by adding sterile phosphate buffered saline (PBS,

pH 7.2). Repeated thawing and freezing should be avoided. Working dilutions should be

stored at +4°C, not refrozen, and preferably used t he same day. If a slight precipitation occurs

upon storage, this should be removed by centrifugation. It will not affect the performance of

the product.
